RHOEO LEAF EXPERIMENT

AIM- To observe GUARD CELLS and EPIDERMAL CELLS in a Rhoeo leaf

APPARATUS REQUIRED-

• Rhoeo leaf
• Watch glass
• Weight
• Dropper
• Blade
• Safranin
• Cover slip
• Slide

PROCEDURE-

1. First of all using the weight crush the leaf from middle in order to break its upper lamina
2. Now using a blade cut a small portion of the crushed tissue
3. Put some water in the watch glass
4. Put that cut transparent portion of leaf into this watch glass using forceps
5. Put two drops of safranin into this watch glass to stain the specimen
6. Now put the specimen carefully on the slide
7. Put the coverslip onto it
8. Put the prepared slide in the microscope and focus it to observe the specimen

OBSERVATION-

1. We will observe the red guard cells on the leaf
2. We will also see red colored epidermal cells.
