First you have to take a small thin layer of its sample ..

then u have to stain it so that all the neccesarry details are clear..

next put tue sample in a slide and observe it through a microscope

Pluck a fresh leaf from a balsam plant.

Fold the leaf and carefully tear along the bruised area of the lower side of the leaf.

We can see a colourless narrow border along the torn edge.

Carefully pull out the thin membranous transparent layer from the lower epidermis using a forceps.

Aim: To prepare a temporary mount of leaf peel to observe stomata.

Materials required: Leaf, watch glass, water, safranin stain glass slide, brush, glycerine, cover slip, compound microscope.

Procedure: Take a freshly plucked leaf. With the help of forceps remove the peel of the leaf from its lower surface. Place the peel in a watch glass containing water. Put safranin stain to the watch glass, so that the peel stains lightly. Slowly take the peel from the watch glass and place on the glass slide with the help of a brush. Put a drop of glycerine on the peel.  Extra stain or water should be removed by soaking with filter paper. Slowly place the cover slip over the peel avoiding air bubbles from going inside. Observe the slide under a compound microscope under both the magnifications.

Observation: We observe epidermal cells that are irregular in shape and no intercellular spaces. Many small pores like structures are seen scattered on the epidermal cells. Each pore contains two bean shaped guard cells which contains chloroplasts and nucleus.

Result: Stomata are present on the lower epidermis of the leaf.
