transparent leaf peel?

1) put it in boiling water and baking soda
2) then with a soft brush carefully clean all the cells of it

The Rheo or lily leaf is taken and at first folded and then torn around the bruised area ranging from the lower portion(dorsal surface) of the leaf.

A colourless thin border is observed near the torn edge of the leaf.

A thin membrane containing transparent layer of the leaf is pulled out from the lower epidermis layer.

This transparent leaf peel is then kept in a petri dish containing water.

In this way , we obtain the transparent leaf peel that can be mounted on a fresh slide , covered with a cover slip and observed under the microscope.
