  • So, on spraying the locating agent which is a solution of ninhydrin on the dried chromatography paper then the solution of ninhydrin reacts with amino acids to form coloured spots by producing a purple compound.

Hence, Coloured spots are formed by the locating agent when they react with amino acids.
