New membrane is assembled primarily in the smooth endoplasmic reticulum. The components (phospholipids) get synthesized by cytosolic enzymes, although some final tweaks involving orientation occur after assembly. Membrane-embedded proteins generally get synthesized by rough ER (the ribosomes attach to the membrane and thread the protein through it). There are other complications involving adding and subtracting membranes as vesicles fuse and bud off, but the base synthesis start in the SER.
