The onion bulb is a modification of the leaves. Onions are made up of multiple layers that are separated by thin membranes.
- Onion cell observation procedure:
- Add a drop of water to the microscopic slide's centre.
- Remove a thin membrane from the onion layer and place it in the middle of the microscopic slide (the drop of water will help flatten the membrane)
- On the onion membrane, place a drop of iodine solution (or methylene blue)
- Place a tiny coverslip on top of the membrane and gently push it down with a needle to remove any air bubbles.
- To drain excess iodine/water solution, place a blotting paper on one side of the slide
- To observe, place the slide on the microscope stage at low magnification.
- To notice clearly, adjust your focus.
- Major observations:
- Cells are large, rectangular, and interlock.
- The cells are surrounded by distinct cell walls that are clearly visible.
- The nucleus is stained darkly.
- In the middle, there are large vacuoles.
- Inside the cells, little granules may be seen (within the cytoplasm).
