If A⃗ and B⃗ are the 2 vectors in question, then the resultant:
R =A⃗ +B⃗
if S⃗ is perpendicular to one of the initial vectors, say A⃗ ,
R .A⃗ =0
→A⃗ .(A⃗ +B⃗ )=0
→A2+ABcos(θ)=0
→θ=cos−1(−A2AB)
→θ=cos−1(−AB)
