A chromatography column with a length of 10.3 cm and inner diameter of 4.61 mm is packed with a stationary phase that occupies 61.0% of the volume. If the volume flow rate is 1.13 mL/min, find the linear flow rate.

Solution:-

The inner volume of the column is πr ²L

V = π × 0.231²cm² × 10.3cm = 1.72cm³

Mobile phase volume = 1.72cm³ × 0.39 = 0.670cm³

The fraction of the column that is swept by the mobile phase each minute

= 1.13cm³min⁻¹ / 0.670cm³ = 1.69min⁻¹

So, linear flow rate = 1.69min⁻¹ × 10.3cm = 17.4cmmin⁻¹
