hello,

I want to measure the size of an rectangular object taken by microscopy. It seams very simple by using imgeJ. However, I made some confusion about the magnification.

In the image below, If I multiply the distance measured by scale bar by the magnification, It does not give the correct answer.

How can I calculate the actual size of this rectangular shape.
