Temporary mount of a leaf peel:

When a specimen is placed in slide with a drop of water is called temporary mount or wet mount. Here, a leaf peel undergoes temporary mount to view the structure of stomata.

Steps for experiment:

  1. Remove a leaf from a plant.
  2. Use forceps to remove a peel from the lower surface of the leaf.
  3. Please the peel in a watch glass containing water.
  4. Put few drops of safranin stain in watch glass.
  5. Keep the peel in water for few minutes.
  6. Take the peel out and place it in the glass slide.
  7. Put a drop of glycerine over the peel with the help of dropper.
  8. Cover the sample with coverslip without any bubbles.
  9. Use blotting paper to remove the excess glycerine and stain.
  10. Place the slide under microscope to view the stomata.

Observations:

Tiny holes appearing in the specimen are stomata.
