Temporary mount of a leaf peel:
When a specimen is placed in slide with a drop of water is called temporary mount or wet mount. Here, a leaf peel undergoes temporary mount to view the structure of stomata.
Steps for experiment:
- Remove a leaf from a plant.
- Use forceps to remove a peel from the lower surface of the leaf.
- Please the peel in a watch glass containing water.
- Put few drops of safranin stain in watch glass.
- Keep the peel in water for few minutes.
- Take the peel out and place it in the glass slide.
- Put a drop of glycerine over the peel with the help of dropper.
- Cover the sample with coverslip without any bubbles.
- Use blotting paper to remove the excess glycerine and stain.
- Place the slide under microscope to view the stomata.
Observations:
Tiny holes appearing in the specimen are stomata.
