If you keep stomata in darkness the transpiration will not take place. Another important thing is that photosynthesis will not take place due to the absence of light

When the Sun goes down, the processes that flora use to generate, produce and store energy slow down considerably or stop altogether. Stomata remain open during daytime to allow for respiration so that these processes can continue, using CO2 from the atmosphere and returning O2 and water vapor as waste products. If the stomata were closed during the day these processes would be retarded severely by the lack of access to CO2 and the build-up of O2. Since the plants, use sunlight and chlorophyll to accomplish this, at night the processes are subdued to the point where stomata no longer need to be open because the chemical factory is basically closed for the night!
