it doesn't matter how tightly you tie knot because it will always have some space between them which may be very very small..But the size of air molecules is smaller than gap so some molecules keep on coming out of the balloon eventually deflates it.
The helium air balloon deflates faster than ordinary balloon because helium is a noble gas it consist of only one atom where is most of the other gases exist as diatomic molecule. So, the size of molecule of helium gas is much smaller than other gas. THEREFORE, the helium gas balloon deflates faster.

Answer:
The air inside the balloon constantly exerts a pressure on the thin, stretched walls of the balloon and keeps on diffusing out gradually through it.
