negative
Can be decolourized to accept counter stain (Safranin or Fuchsine); stain red or pink, they don't retain the Gram stain when washed with absolute alcohol and acetone.
positive
Retain crystal violet dye and stain dark violet or purple, they remain coloured blue or purple with gram stain when washed with absolute alcohol and water.

GRAM POSITIVE BACTERIA:
1.Cell wall made up of peptidoglycan+teichoic acid.
2.Stain remain persistant.
3.Cell wall single layered.
4.Only 2 rings in flagella present — S and M.
5.Very low lipid content.
GRAM NEGATIVE BACTERIA:
1.Cell wall made up of peptidoglycan.
2.Statin removes because stain is soluble in water and insoluble in lipid.
3.An additional layer of glycolipid is present in the cell wall.
4.Teichoic acid absent.
5.All 4 rings in flagella present — L,P,S and M.
High lipid content.
