In DNA fingerprinting- It is employed in the last steps to autoradiograph the fragments in which the fragments are exposed to DNA probes or pieces of synthetic radioactive DNA that are bound to the minisatellites. Fragments are exposed to the X-ray film and a dark mark was produced at any point where the radioactive probe had attached itself. The resultant pattern is created by the marks.
2. In Southern hybridization- DNA fragments are denatured in situ and separated on agarose gels, the fragments are then transferred to a solid support and are immobilized. To reduce the non-specific hybridization, they are prehybridized with a probe, and hybridization of the filter or the membrane takes place according to the desired radiolabelled nucleic acid probe. To remove the unbound & weakly binding probe, the filter or the membrane is washed and then autoradiographed.
