Plastid transformation is typically performed by either biolistic bombardment of plant tissue with a transformation vector (Svab and Maliga, 1993) or by polyethylene glycol-mediated transformation of protoplasts (Golds et al., 1993). Plastid transformation is achieved by homologous recombination between the transformation vector and the plastid genome, resulting in integration of the gene(s) of interest at a predictable, pre-determined site (Maliga, 2004). The most effective selectable marker has been aadA, which encodes aminoglycoside adenyltransferase and confers spectinomycin resistance (Day and Goldschmidt-Clermont, 2011; Maliga, 2004).
