Firstly you should note that CO2 is transported as negative HCO3 ions. Dissolved CO2 reacts with water to form carbonic acid which happens spontaneously in the plasma but is accelerated by a zinc containing enzyme known as the carbonic anhydrase in the RBCs. The carbonic acid thus formed dissociates immediately, yielding H+ ions and HCO3- ions. H+ then combines with hemoglobin present in the RBCs. The HCO3 ions exit into the plasma which creates an Ionic imbalance. Thus Cl- ions diffuse from the plasma into the RBCs. This is also know as the Chloride shift/Hamburger’s Phenomenon.
