These enzymes catalyse covalent bonding of two substances to form a large molecule. they catalyse joining of C - O, C- S , C- N , P-O etc. bonds by using energy ATP.

Example: RUBP carboxylase , ligase, phosphoenol pyruvate (PEP Carboxylase) etc.

Phosphoenol pyruvic acid + CO2──> oxaloacetic acid
