GRAM POSITIVE:Gram-positive bacteria retain the color of the crystal violet stain in the Gram stain. This is characteristic of bacteria that have a cell wall composed of a thick layer of a particular substance (called peptidologlycan).

GRAM NEGATIVE:Bacteria whose cell walls are made up of an outer membrane in addition to a inner peptidoglycan layer are called as gram negative.These bacteria get decolorised by alcohol owing to the lipidic outer membrane and lose the stain crystal violet, so they have to be counterstained by secondary stain safranin appear pink in color.
