  • It's in vitro technique used for amplification of nucleic acids.

Steps in polymerase chain reaction -

  • Denaturing stage - 94-95 ℃
  • Annealing stage - 50-56 ℃
  • Extending stage - 72 ℃

Denaturation of desired DNA into two strands, each acting as a template, for each strand separate set of primer ( two sets of primer) used, with the help of deoxy(ribo)nucleotides and Taq polymerase (DNA polymerase isolated from Thermus aquaticus ), extension of DNA template occurs, resulting in replication of desired DNA (amplification) = ½ × 6
