Describe how chromatin gets assembled
Answers
In vivo studies have indicated that the DNA replication machinery (large red oval) displaces all the histones from the parental chromatin. The two daughter DNA duplexes are rapidly assembled into chro- matin in a stepwise manner from a mixture of old histones and de novo synthesized histones.
Chromatin Assembly and the diversity of chromatin network takes place through multiple steps:
Step by step explanation:
1) The assembly initiates with the deposition of histone H3-H4 tetramer on DNA.
2) This is followed by the addition of two histone H2A- H2B dimers to core particles.
3) The addition of the linker histones is accompanied by folding of the nucleofilament.
4) During the initial assembly, the incorporation of structurally modified histones, produces an array of structural modified relationships.
5) This is carried out through numerous ways like methylation, phosphorylation, acetylation etc. This therefore, results in modified chromatin structural activity.
6) In the maturation phase, differential modifier histones, for instance, HMG proteins and other DNA binding factors aids in the unfolding and the modification of the nucleosomal array. This further followed by histone modification.
7) Stimulatory factors bind histones to several domains, thereby enforcing remodeling of the structural and functional activities. Moreover, the incorporation of histone- interacting factors further produce specificity to the chromatin assembly.
