Proteins are detected using Biurette reagent.
This turns a mauve or purple colour when mixed with protein.

Protein in the food can be tested using any of the following methods: Biuret test, Ninhydrin test, Xanthroproteic test, or Millon's test.
- 1) Biuret test
- prepare a food extract
- Add Aqueous copper sulfate.
- If the solution turns violet, it indicates the presence of protein.
- 2) Ninhydrin test
- Prepare the food extract.
- Add pyridine ninhydrine solution.
- If the solution turns violet, it indicates the presence of protein.
- 3) Xanthoproteic test
- Prepare the food extract
- Add nitric acid solution.
- If the solution turns yellow, it indicates the presence of protein.
- 4) Millons test
- Prepare the food extract
- Add sodium nitrite and sulphuric acid to it followed by mercuric sulfate
- Brick red color appears if there is a protein
