Formation of recombinant DNA is described below-
To form a recombinant DNA, first of all, you require a vector. A vector is any molecule which carries desired foreign genetic material or gene of interest in the host organism.
Host organism is the organism whose gene has to be modified. Vector can be plasmids, bacteriophage etc.
Now, vectors have restriction endonucleases sites. These are specific sites which can be cleaved only by their particular restriction endonuclease enzymes.
Example EcoRI site on a vector can only be cleaved by EcoRI restriction endonuclease enzyme. The gap thus formed in vector, is filled by the gene of interest to be inserted and joined by ligase enzymes.
The vector is further inserted into the host organism through different methods like electroporation, transformation etc. EcoRI site is - 5'-GAATTC-3'.
