- The clotting system consists of a mixture of components including cells, fragments thereof, zymogens, their activation products and naturally occurring inhibitors, adhesive and structural proteins, phospholipids, lipids, cyclic and noncyclic nucleotides, hormones and inorganic cations, all of which are normally maintained in a state conducive to blood flow.
- Upon damage to the vascular wall with disruption of the monocellular layer of endothelial cells, exposing the subendothelial layer, and also permitting bleeding, a series of events is initiated designed to produce clot formation.
- These homeostatic reactions lead to the generation of the primary platelet plug, followed by a clot principally containing cross-linked fibrin (secondary hemostasis).
- Subsequently, the blood vessel is repaired and the clot is dissolved by a mechanism which includes fibrinolysis.
