Using the recombinant DNA technology it is possible to isolate and clone single copy of a gene or a DNA segment into an indefinite number of identical copies. So that the inserted DNA also replicates faithfully with the parent DNA is called as gene cloning.

  1. A fragment of DNA to be cloned is first inserted into a plasmid vector containing an amipicillin-resistance gene(amp), shown in given figure.
  2. Only the few cells transformed by incorporation of a plasmid molecule will survive on ampicillin-containing medium.
  3. In transformed cells, the plasmid DNA replicates and segregates into daughter cells, resulting in formation of an ampicillin-resistant colony.
