Tonoplast can be defined as membrane of vacuoles that separate contents of vacuole from cytoplasm of the cell. It is also known as vacuolar membrane and involved in regulation of ion movements across the vacuoles.

Plasma membrane is an outer covering of the cells that separates cytoplasm of the cell from its external environment. As tonoplast, plasma membrane also regulates transport of ions and other substances.

Thus, tonoplast is outer covering of vacuole, while plasma membrane is outer covering of cells.
