Decalcification describes the technique for removing mineral from bone or other calcified tissue so that good-quality paraffin sections can be prepared that will preserve all the essential microscopic elements. Decalcification is carried out after the specimen has been thoroughly fixed and prior to routine processing to paraffin. In this paper the basic structure of bone is described and the technical options for preparing sections are outlined. The procedure for decalcification and the successful monitoring of the process is discussed and some popular options for the choice of reagents are provided.
