There is no difference between them .
both are same .

examples :He was very fast in spite of being terribly overweight.

They arrived late despite leaving in plenty of time.

the only difference between inspite and despite is the 'of'. Despite the pain in his leg he the marathon. Inspite is a slightly more elaborate form , to be used in more fancy , most formal writing. Despite is not informal but not quit elaborate.
