If the gene is inserted using Eco RI restriction enzyme than, tetracycline resistance property of plasmid will be lost due to insertional inactivation if tetracycline resistance gene because Site for Eco RI is present inside tetracycline resistance gene. Desired gene is inserted inside tetracycline gene, tetracycline gene is disrupted which is called as insertional inactivation of tetracycline gene, as now property of tetracycline resistance is lost. But kenamycin resistance gene is still intact. In first step transformants and selected from non-tansformant by plating on kenamycin containing medium as transformant have intact kenamycin gene (due to presence of hypothetical plasmid inside host cell) but non-transformant fails to grow on kenamycin containing medium due to lack of kenamycin resistance gene (Hypothetical plasmid is not present inside host). Recombinant and non-recombinant are selected from transformant based on tetracycline resistance gene. Non-recombinants will have intact tetracycline resistance gene as no gene was inserted in them and they do not show insertional inactivation of tetracycline resistance gene. Recombinants will not grow on tetracycline due to insertional inactivation of tetracycline gene. Thus kenamycin is used for selection of transformants and tetracycline is used for selection of recombinants. Answer is option (1).
