A polynucleotide strand is made up of nucleotide polymers. Each nucleotide is made up of a five carbon, pentose sugar, a phosphate group, and a nitrogen base. The nucleotide molecules join together via phosphodiester bond between hydroxyl group of one nucleotide and phosphate group of another nucleotide.

Each DNA strand show polarity and gives a 5' and a 3' end. The 5' end of DNA contains a phosphate group, while the 3' end contains hydroxyl group (-OH group).
