  • pBR322 is a plasmid which is used for the purpose of cloning foreign DNA fragments.
  • The plasmid vector is replicated in host bacteria, Escheresia coli or E.coli .
  • It consists of the following parts:
  1. Restriction sites with specific DNA sequences which are recognised by specific restriction endonucleases and are the location where the foreign DNA is introduced in the vector.
  2. An origin of replication called Ori where the replication of the plasmid DNA starts.
  3. A Rop gene helps in maintaining the plasmid copy number in the host cell. This is necessary to prevent the foreign DNA sequences from recombining with each other.
  4. A gene encoding for antibiotic resistance, Ampicillin resistance gene helps to specifically select the plasmid transformed bacteria from the non-transformed ones.
