We know that,
Force on a charge (F) in Electric Field (E) is given by
F = qE -------------- (i)
Now, To place this charge in 'E' you will have to do some work against this Force 'F'.
Therefore, Small work done to move a charge through Potential Difference (V) is
dW = - F.dr -------------- (ii)
From (i) & (ii) we get,
dW = q.E.dr
But dW = q.dV ( For moving a charge through Potential 'V' )
Therefore, q.dV = - F.dr = - q.E.dr
Hence, E = -dV/dr
