these lines were said by Caesar when Brutus whom Caesar considered to be his most loyal friend stabbed him.

what exactly this lines means is that Caesar is meaning to say you too Brutus because Caesar could not believe his friend had done this to him , further more Caesar could have withstood the stab of all the other conspirators but the stab of Brutus was the unkindest of all and Caesar could not accept it and dies.

the conspirators surrounded Caesar in the senate and started stabbing one by one. Casca was the first to strike followed by Casius and others. Brutus was the last to stab Casear. When Casear saw Brutus stabbing his eyes couldn't believe him. He was totally shocked and uttered in surprise, 'Et tu Brute'. Brutus was Caesar's 'Angel'. Seeing this, Caesar fell down dead without making any Resistance.
