Explanation:  

  • Heamatoxylin is a basic stain. Basic stain reacts with acidic substance and vice versa. Now the nucleus of the cell consists of nucleic acid.  
  • This nucleic acid being acidic in nature gets stained by the haematoxylin stain. The stain produces a characteristic purple colour on reaction with the acid.  
  • Haematoxylin is used along with eosin which stains the basic component of the cell. Heamatoxylin and eosin are used in the biopsy slide study.
