Onion Cell:

  • The outer Epidermal layer of Onion acts as a protective layer and protects the inner soft tissues from Bacteria, Virus and Fungus.
  • Because it has a simple cell structure and are transparent, It is used to show students and explain about plant cells.

Cheek Cell Experiment:

1. Take a small clean cotton ball. Gently brush the inside of your mouth with the cotton ball.

2. Take a slide that is used to keep under a Compound Microscope. Stain its center with the cotton ball that you brushed the inside of your mouth with.

3. Add a drop of Methylene-blue solution to this. This solution colours the negatively charged molecules in the cell, like DNA and RNA and makes the cell components to be clearly visible.

4. Place a cover-slip on top of this and keep this slide under a 4× or 10× Compound Microscope. If Methylene blue solution is added in excess, the cover-slip makes the excess solution to come to the edge, where you can remove through a paper towel.

5. When you view the slide under Higher Magnification, you can see 'Squamous Epithelial Cells.' These cells are only one-layer thick. A dark blue coloured center Nucleus with small blue dots that are bacteria present in your mouth are also clearly visible.
