I Am Writting Full Experiment as i have done it .........

.AIM ...=========to prepare a temporary mount of leaf peel to show its stomata .....

.Requirements ...================Leaf of a plant like lily, Bryophyllum , or tradenscantia, forceps, waych glass, slide, coverslip , brush, needles, safranin, glycerine, and microscope .

Principal Involved ...===================Stomata are minute appertures ( openings ) present on the surface of the leaf . they are generally more on the lower epedermis . Each stomata is surrounted by two bean-shaped cells called “gaurd cells” . Theae cells have differential thickenings, i.e. thicker inner wall facing the stoma and thinner outer wall . The turgidity of gaurd cells controls the opening and closing of stomata . Stomata heps in exchange of gases and water vapours between the atmosphere land leaf .

Procedure ...===============

1 ) Pell off the leaf from its under surface and put the peel in a watch glass containing water .

2 ) Add a few drops of Safranin stain into the watch glass to stain the peel

.3 ) After staining put the peel on the cleaning slide and cut it in a rectangular or square shape .

4 ) With the help of a filter paper, remove the excess of stain and water

.5 ) Put a drop of glycerine on the slide over the peel, gently put the coverslip to avoid the entry of air bubbles and observe under the microscope .

Observations ...=================

1 ) Epidermis consists of a single layer of cells which are irregular in outline and without intracellular spaces .

2 ) Minute appertures ( openings ) are seen which are imbedded in epidermal cells

The minute apperture seen in the mount are stomata . Each stomata is surrounded by two bean-shaped cells called gaurd cells

.Precautions ...================

1 ) The epidermal peel should be taken from a freshly plucked leaf

.2 ) Always hold the slide by its edges to avoid making the slide dirty .

3 ) Always use a brush to transfer the peel from petri dish to the slide

4 ) The peel should be cut to a proper size and its curling must be avoided

.5 ) The peel should be mounted in the centre of the slide .

6 ) The peel should not be allowed to dry .

7 ) Always keep the coverslip gently to avoid the entry of air bubbles .

8 ) Oozing of glycerine should not be there .
