Gene amplification is a stage wherein several copies of a single gene is made without affecting the other genes. This could be a natural process or an artificial one.

The annealing stage is the stage at which two strands of DNA from a single strand are formed and rejoin.  This will only happen when the temperature drops or returns to the natural level.
