Explain principle and application of immunoelectron microscopy.
Answers
Definition and application:
●it can be defined as any technique that uses antibodies, or molecules that interact with antibodies in conjunction with electron microscopy to localize ultrastructurally antigens or antibodies in cells and tissues. In addition, a number of other biological macromolecules whose specific ligand-bind-ing properties are known can be used.
Principle:
●The principles of two representative methods, pre-embedding and post-embedding methods, of immunoelectron microscopy were explained, and their merits and demerits in application for clinical medicine were described. Some examples of the preembedding method were demonstrated with the immunoelectron micrographs. The usefulness of Laser Scanning Microscope in visualizing subcellular localizations of antigens was also discussed.
