Explanation:

Chemical compostion may be basic, acidic, amphoteric (neutral). According to these characters stain different components of tissue. These are cationic dyes and stain nuclei, basophilic granules or bacteria. These are anionic dyes and stain mainly cytoplasm, eosinophilic granules.
