The simplest experiment to demonstrate positive geotropism of roots is placing seedlings in the dark. The seed on germination develops a radicle and a plumule oriented towards gravity and light respectively. When the seedling in dark is rotated by 180 (upside down) or 90 (placed on its side) the root will bend and grow downwards, negative phototropism can be ruled out since the plant is not exposed to any source of light. Roots in absence of light grow downwards, clearly shows that root cells can perceive gravity with the help of specialized cells called Statocytes or amyloplasts or in simple words starch grains. Under the influence of gravity these starch grains move towards the cell (statocyte) membrane, depending on where the starch grains fall, where these grains touch the membrane the way down is determined. These statocytes in the root cap then send these signals (transduction) to the root tip (the meristematic cells) which is responsible for the directional growth of the root.
