ANSWER:-

Let the medium thickness be t, then the right edge is at x = t/2 and left end is at -t/2.

To obtain the transmission coefficient, T the ratio should be of the amplitudes of after traversing and before entering. so T = Amp(x = -t/2) / Amp(x = +t/2).

And, reflection coefficient would be simply, R = 1 - T (assuming there aren't any other type of propagation involved).

If the wave is undergoing losses in the medium before it hits the target solid medium, then you will have to account for the losses too if you wish to calculate R by the methods of amplitude ratios.
