Initiation: tissue factor (TF) is exposed by vessel damage; coagulation factor VIIa (FVIIa) binds to TF and activates small amounts of FX to FXa and FIX to FIXa. This leads to trace amounts of thrombin. Amplification: the trace amounts of thrombin activate the co-factors FV and FVIII to FVa and FVIIIa and platelets (via cleavage of protease-activated receptors, PAR1 and PAR4), which leads to the formation of complexes of factors for more efficient thrombin generation
