Preparations of Rous sarcoma virus, purified by density gradient centrifugation, have been examined by electron microscopy and chemical analysis. Sections of the pellet obtained by high speed centrifugation of such preparations show a high degree of homogeneity. Examination of shadowed preparations, combined with virus assays, allowed an estimate to be made of the number of physical particles per infectious unit. This number was 750 for tissue culture fluid and 1200 for purified preparations.
