HOFFMAN'S MUSTARD OIL TEST:-

When alkyl amine is heated with carbon disulfide and mercuric chloride alkyl isothiocyanate is formed which has a smell like mustard oil and hence it is known as Hoffman mustard oil test.

Only one degree alkyl amine give this test.

R-NH2 + S=C=S ----->R-NH-C=S ---->R-N=C=S+HgS +2HCl

R2-NH + S=C=S ------->no reaction

R3-N +S=C=S---------->No reaction

Hoffman mustard oil reaction is used to test primary (1°) amine.

Explanation:

This  reaction include primary amine react with phosgene (CS2 ) or carbonyl chloride (CoCl2) , by heating we get dithioalkyl carbamic acid , after this when the product react with mercuric chloride (HgCl2 ) it will form alkyl isothiocynate , smell like mustard oil so the name of reaction is " Hoffman Mustard Oil Reaction ".

       RNH2  +    S- C-S +  Δ   →  R- NH-C-S-H

        R-NH-C-S-H   + HgCl2 →   R-N= C=S  +   HgS  +  HCl
