In my experience, we can develop a PCR primer set or more to amplify mutation genes.

1- we collect the nucleotide sequence of both original gene and mutation genes.

2- decide the primer at that point of mutation. So that designed primer would be able to amplify only the gene that get mutated. We also decided a primer set for amplify the same position to the original gene.

3- There should be 1 positive control, for 1 positive original gene and mutation genes

4- i was success to use in my study, but i recommend you to consult with other molecular biologist
