As much I know the reaction take place only when the spern needs to penetrative ovum wall.

During fertilisation a sperm fuses with the plasma membrane and then penetrate the female egg in order to fertilize it. Fusion with the egg causes problems and fusion is a slow process. The penetrating through the egg's hard shell or extracellular matrix which is present more of a problem to the sperm. As the sperm approaches the zona pellucida of the egg it is necessary for initiating the acrosome reaction in the sperm cell.
