Hey

I don’t think there’s a formula. But phosphodiester bond is between PO4- group and sugar. You can do one thing for calculating it

Suppose your DNA is 50bp long.

That means it has 50 bases each strand. And, each base is connected to sugar. Each sugar is bonded to other by phosphodiester bond except the first one (5′ end) and last one ( no phosphate justOH 3′), So the number of phosphodiester bond is 49. (Thanks harman for pointing it out)

Don’t forget doubling it because 50bp DNA means double stranded DNA.

So, total Phosphodiester Bond - 49+49= 98.
