Pancreatic juice contains the proenzymes trypsinogen, chymotrypsinogen, procarboxypeptidases, and proelastase. All are activated by trypsin in the intestinal lumen. Enteropeptidase located in the brush border of the jejunal mucosa converts trypsinogen to trypsin.

Pancreatic trypsinogen is activated by duodenal enterokinase to become trypsin. Trypsin then acts on zymogens to form more trypsin (autocatalysis), chymotrypsin, and carboxypeptidases. α-Amylase is secreted in an active form, as are lipases, ribonuclease, and deoxyribonuclease
