Depends on which country you are from and which government you mean to serve. If you are an Afghan who wants to serve the Americans to help them assassinate Taliban leaders the rules are very different from if you want to work for your own government.
In the former case all you have to prove is you have unique access to high value targets and sufficient motivation to carry through the attack.
In the latter case you would typically need conventional training to start off, become enlisted or commissioned in your country’s military, rise through the ranks toward either intelligence or front line combat roles. Attend your military’s combat and skill proficiency schools, move up the tiers of specialized direct action units until you reach what is commonly referred to as “Tier 1” operations (this is not actually a real term officially but many people use it).
At which point exceptional individuals are recommended by their command structure for the possibility of joining what you might be referring to as “assassination” programs. These programs don’t actually exist, in that there is no such thing as unit or department that only does assassinations as far as anyone publicly knows. It is more likely that a intelligence operative spends most of his time gathering human intelligence or developing human assets than killing anyone. They are also more likely used to help train or organize assets to conduct the killing than actually doing it themselves.
In the real world a regular disgruntled janitor could be more effective at killing the leader of a nation than a super spy trained by the government. The cost of failure is also lower if the operation fails or is exposed during the planning stages. It is also much easier to deny the involvement of your own government if it is a local who attempted the assassination rather than a government trained operative.
