to examine the starch accumulation in your experimentally treated leaves ,you first need to remove green chlorphyll from them by usinggreen leaftest tubealcoholcontainerwaterprocedure:
place leaf in a test tube and pour some alcohol in it. Heat some water in a container and put test tube containing alcohol and green leaf into the container.chlorophyll dissolves in warm alcohol .Thus,leaf looses it green colour and become decolourise
