In macroscopic cases,you can do a lot of tests like,
1- Take a paper and try to pass it between the gap(if exists it will pass,otherwise not)
2-Put the system in water and if space is there bubble will be formed.
3- Take a thin thread and try the first method.
In microscopic level-
1- Compound Microscopy.
2- You can use friction as a perimeter to know the space too.
